Adsorption of Ethidium Bromide with activated charcoal:

1. pre-packaged filter units:

are made by Schleicher and Schuell (S&S Extractor™)

  • available from Mandel Scientific for about CDN $20 each or Sigma Chemical Company (CDN $27.10 each).
  • able to absorb at least 5 mg of EB in solution containing 0.5 µg/mL (capacity of at least 10L according to instructions)
  • reduces EB to less than 0.8% of original concentration (spectroscopic assay)
  • after filtration, the solution can be flushed down the drain
  • the charcoal filter is bagged, tagged and disposed of by EH&S

2. charcoal absorption in bulk solution: (reference 1)

  • dilute EB solution to less than 10 µg/mL
  • for a volume of 100 mL (1 mg EB)
  • add 300 mg of activated charcoal
  • stir for 20h at room temperature
  • recover charcoal containing EB by filtration, flush filtrate down the drain
  • put filter paper and charcoal containing EB into a plastic bag for disposal by EH&S
  • expect EB concentration in filtrate to be less than 0.05 µg/mL (<0.5% original concentration) if inintiall solution is less than 10 µg/mL
  • system effective with EB in water or buffer solution (MOPS or TBE) or saturating concentration of cesium chloride.· reference suggests that adsorption may be complete in an hour, but for safety, leave for 20h before filtering.
